SacredBod's longer take on Pregnenolone — context the structured blocks above don't capture.
What It Is
Pregnenolone is a C21 steroid and the biochemical precursor to all other steroid hormones — including progesterone, cortisol, aldosterone, DHEA, testosterone and estradiol. Synthesised from cholesterol in the mitochondria, it is also classified as a neurosteroid because it is produced de novo in the brain and modulates neuronal excitability. Pregnenolone and its sulfate ester (PREG-S) act as positive allosteric modulators of NMDA receptors and negative modulators of GABA-A receptors, influencing memory, mood and neuroplasticity.
How It Works
Pregnenolone sulfate enhances NMDA receptor function and long-term potentiation (LTP), the cellular basis of memory. It increases acetylcholine release in the hippocampus and cortex, supporting cholinergic neurotransmission. In DAT knockout mice (a model of ADHD and mania), pregnenolone normalised hyperactivity and reduced impulsivity. It also stimulates microglial autophagy, reduces neuroinflammation and promotes remyelination in models of multiple sclerosis. Low pregnenolone levels are associated with cognitive decline in ageing and bipolar disorder.
Who Should Consider It
Adults experiencing mild cognitive decline, brain fog or low mental clarity. Those with age-related memory concerns. Some individuals with mood disorders (bipolar depression) have been studied under psychiatric supervision. Athletes and biohackers interested in neurosteroid optimisation. Not a replacement for hormone replacement therapy.
Dosage Guide
Typical dose: 10 mg per day
Form: capsules (180 count)
Best time: morning
With food: with-food
Expected onset: 2–4 weeks for mood; 4–8 weeks for cognitive benefits
Cycling: Some practitioners recommend cycling (3 weeks on, 1 week off) to prevent receptor downregulation, though evidence is limited.
Safety & Side Effects
Known side effects: Generally well-tolerated at 10–30 mg/day. Higher doses may cause irritability, anxiety, insomnia or acne (androgenic conversion). May cause headache or dizziness in sensitive individuals. Rare reports of heart palpitations at high doses.
Who should avoid: Pregnant and breastfeeding women. Individuals with hormone-sensitive cancers (breast, prostate, ovarian). Those with bipolar disorder or seizure disorders (neurosteroid modulation may destabilise mood or lower seizure threshold). Children and adolescents. Not for use as a steroid hormone replacement without endocrinological supervision.
Avoid combining with: Hormone replacement therapy, DHEA supplements (redundant steroid precursor stack), Corticosteroids, Anticonvulsants, Benzodiazepines (theoretical GABA-A interaction), SSRIs (theoretical serotonergic interaction)
India-Specific Context
Pregnenolone is available on Amazon India with varying brand quality. When selecting a product, verify standardization claims against the evidence base cited above. Indian brand preferences include Carbamide Forte, HealthyHey, Nutrabay Pure, Pure Nutrition, Now Foods, Nutricost, Himalaya, Patanjali, Dabur, Trexgenics, Evorina, Nervana, and Life Extension. Prices vary significantly; compare cost-per-active-dose rather than capsule count alone.
Schedule status in India: Not a Schedule H drug; available as dietary supplement/herbal product.
